== Plot == After the alien invasion at the Black Mesa Research Facility, a multidimensional alien empire known as the Combine has conquered Earth. Approximately twenty years after being placed in stasis, Gordon Freeman is inserted into a train bound for City 17 by the G-Man (Michael Shapiro). Helped by the undercover Resistance member Barney Calhoun (Shapiro), Gordon attempts to reach the laboratory of Dr. Isaac Kleiner (Harry S. Robins) but is subdued by Combine officers. He is rescued by Alyx Vance (Merle Dandridge), who guides him to the laboratory. Kleiner's attempt to teleport Gordon to the Resistance base fails, and Gordon is momentarily teleported to the Citadel, the skyscraper headquarters of Dr. Wallace Breen (Robert Culp), the former Black Mesa administrator and the Combine's puppet ruler. Gordon uses an airboat to progress to the base via the city's canal system, eluding Combine forces. At the base, Gordon reunites with Alyx's father, Dr. Eli Vance (Robert Guillaume), and meets Dr. Judith Mossman (Michelle Forbes). Alyx introduces Gordon to her pet robot, Dog, and gives Gordon the gravity gun. Combine forces storm the base and capture Eli and Mossman. Negative Ames test means the possible mutagen DID not cause increase in revertants and positive Ames test signifies that the possible mutagen DID increase the chance of mutation. These mutagenic effects on bacteria are researched as a possible indicator of the same effects on larger organisms, like humans. It is suggested that if a mutation can arise in bacterial DNA under presence of a mutagen then the same effect would occur for larger organisms causing cancer. A negative Ames test result could suggest that the substance is not a mutagen and would not cause tumor formation in living organisms. However only few of the positive Ames Test resulting chemicals were considered insignificant when tested in larger organisms but the positive Ames test for bacteria still could not be conclusively linked to expression of cancer in larger organisms. === Zeolites === Zeolites are natural or synthetic crystalline aluminosilicates, which have a repeating pore network and release water at high temperature. Zeolites are polar in nature. They are manufactured by hydrothermal synthesis of sodium aluminosilicate or another silica source in an autoclave followed by ion exchange with certain cations (Na+, Li+, Ca2+, K+, NH4+). The channel diameter of zeolite cages usually ranges from 2 to 9 Å. The ion exchange process is followed by drying of the crystals, which can be pelletized with a binder to form macroporous pellets. Zeolites are applied in drying of process air, CO2 removal from natural gas, CO removal from reforming gas, air separation, catalytic cracking, and catalytic synthesis and reforming. Non-polar (siliceous) zeolites are synthesized from aluminum-free silica sources or by dealumination of aluminum-containing zeolites. The dealumination process is done by treating the zeolite with steam at elevated temperatures, typically greater than 500 °C (930 °F). This high-temperature heat treatment breaks the aluminum-oxygen bonds and the aluminum atom is expelled from the zeolite framework.
=== 13 April === The Ukrainian Defence Ministry confirmed that Russian forces had reached the northern outskirts of Bohdanivka, ten kilometers from Chasiv Yar. Russia claimed to have taken Pervomaiske, 11 kilometers west of Avdiivka. Three people were killed in separate Russian attacks in Donetsk Oblast, while two people were killed in Kharkiv Oblast. One person was killed in a Russian attack on a car carrying humanitarian aid in Chernihiv Oblast. ==== Improved reproducibility ==== Overcoming issues of reproducibility has become a topic of growing concern across scientific disciplines. Reproducibility can be especially salient when multiple iterations of the same experimental protocol need to be repeated. Using liquid handling robots that can minimize volume loss between experimental steps are often used to reduce error rates and improve reproducibility. An automated DMF system for CRISPR-Cas9 genome editing was described by Sinha et al, and was used to culture and genetically modify H1299 lung cancer cells. The authors noted that no variation in knockout efficiencies across loci was observed when cells were cultured on the DMF device, whereas cells cultured in well-plates showed variability in upstream loci knockout efficiencies. This reduction in variability was attributed to culturing on a DMF device being more homogenous and reproducible compared with well plate methods.

==== Increasing resistant starch intake ==== Resistant starch is a type of non-digestible, fermentable fiber resistant to amylase digestion in the small intestine. It is broken down to short-chain fatty acids by microflora in the large intestine. It is commonly found in cooked and cooled potatoes, green bananas, beans and legumes. The short chain fatty acids can lead to further oxidation of fat and a higher energy expenditure. Resistant starch dilutes energy density of food intake, maintains a bulking effect similar to non-fermentable fiber, and increases the expression of gut hormones PYY and GLP-1. The increase in gut hormone expression affects neuronal pathways in the brain that contribute to long-term energy balance and improved overall health of the intestines.
