=== Enzyme induction and inhibition === The duration and intensity of pharmacological action of most lipophilic drugs are determined by the rate they are metabolized to inactive products. The Cytochrome P450 monooxygenase system (CYP) is a crucial pathway in this regard. In general, anything that increases the rate of metabolism (e.g., enzyme induction) of a pharmacologically active metabolite will decrease the duration and intensity of the drug action. The opposite is also true, as in enzyme inhibition. However, in cases where an enzyme is responsible for metabolizing a pro-drug into a drug, enzyme induction can accelerate this conversion and increase drug levels, potentially causing toxicity. For example, chemotherapy prodrugs like cyclophosphamide (CPA) and ifosfamide (Ifex), which are initially inactive, become toxic as they are metabolized into cytotoxic compounds (such as phosphoramide mustard and chloroacetaldehyde) primarily from liver enzymes CYP2B6 and CYP3A4. == History == AZD-2327 was first described by 2004 and was first described in the scientific literature in 2009. The development of AZD-2327 was discontinued in 2010. It reached phase 2 clinical trials for both depressive disorders and anxiety disorders prior to its discontinuation. No reason was given for the discontinuation of its development. However, the drug was found to be ineffective for major depressive disorder in a phase 2 clinical trial. AZD-2327 was developed by AstraZeneca.

== History == Minocycline was a commonly used tetracycline synthesized in Lederle Laboratories in 1970, but antibiotic resistance to the drug began growing in prevalence throughout the 1970s and 1980s. While the problem of antibiotic resistance was known to scientists during the 1980s, apathy led to little federal attention given to the emerging crisis. However, by the late 1980s the worldwide threat began to be treated more seriously, which led to the renewed funding of antibiotic research. In 1993, researchers in the same laboratories that first synthesized minocycline created a new generation of tetracycline antibacterial agents, known as the glycylcyclines. These antibiotics were the first new drugs of the tetracycline class to be reported since the discovery of minocycline in 1970. The glycylcyclines were found to be active against a broad spectrum of tetracycline susceptible and resistant Gram (-) and Gram (+) aerobic and anaerobic bacteria. This initial research resulted in numerous studies being done on the antibacterial activity of various glycylcyclines, with extra focus being put on N,N-dimethylglycyl-amino derivatives, due to their reported potency. Hegseth signed a memo allowing military personnel to carry personal firearms on military installations such as forts, camps, bases, naval yards and recruitment centers. His stated purpose was to allow personnel to defend themselves in the event of an attack and to support their second amendment rights. Supporters of the new policy, such as the family of slain Fort Hood specialist Vanessa Guillen have applauded it saying it would save lives. Concerns remain as to how the new policy will work in practice with state firearm laws, emergency response, and active threats.
== Career == Huttenlocher joined the faculty at the University of Wisconsin–Madison (UW-M) in 1999 with a joint appointment in the Departments of Pediatrics and Pharmacology, and as an associate director of the Medical Scientist Training Program (MSTP). In this role, Huttenlocher's research has defined cellular and molecular mechanisms that regulate cell migration and identified basic adhesive mechanisms that regulate cell migration and leukocyte chemotaxis. By 2005, her research team had identified a novel pathway that turned out to be critical for cell migration and chemotaxis, involving intracellular proteolysis by the calcium-dependent protease calpain. Her laboratory was also the first to document reverse migration of neutrophils away from sites of injury or inflammation. In 2011 her group reported the first redox sensor that mediates leukocyte attraction to wounds. In 2005 she was elected a member of the American Society for Clinical Investigation. Huttenlocher was promoted to Professor with tenure in 2008 and was the recipient of the Graduate School’s H.I. Romnes Fellowship award. She also received the Burroughs-Wellcome Fund’s Clinical Scientist Award in Translational Research for her project "Diagnosis and Treatment of Autoinflammatory Disease."
