In the 1970s Knox and his Edinburgh research group invented new micro-particulate packing materials for liquid chromatography, now marketed under the trade name Hypersil. === Drug testing in the workplace === Workplace drug testing has been widespread and controversial in the US since the late 1980s: there is no clear measure of its effectiveness in improving safety and productivity, and testing affects significantly more non-whites than whites. Most common is urine analysis for amphetamines, cocaine, marijuana, opioids and PCP; usually with no practical discrimination between the effects of the different drugs. Workplace testing rapidly gained popularity after the Reagan administration made it mandatory for federal workers, peaking in 1996, with 81% of companies reporting drug screening, up from 21% in 1987. In the 1980s, testing had been promoted to business as a way to reclaim what were said to be huge losses in productivity caused by drug use. Studies released in the 1990s refuted these claims; a 1994 report from the National Academy of Sciences, "Under the Influence? Drugs and the American Work Force", concluded that "the data... do not provide clear evidence of the deleterious effects of drugs other than alcohol on safety and other job performance indicators." ==== Food and Drug Administration (FDA) ==== The FDA reviews commercially available test kits and their associated medical claims to ensure safety and efficacy before products are cleared or approved for use. The regulatory landscape for laboratory developed tests (LDTs) has been evolving significantly. On May 6, 2024, the FDA issued a final rule significantly modifying its approach to oversight of laboratory-developed tests, amending regulations to clarify that in vitro diagnostic products (IVDs) are devices under the Federal Food, Drug, and Cosmetic Act, including when manufactured by a laboratory. The final rule includes a policy under which FDA would phase out enforcement discretion for LDTs over a four-year period. The phaseout policy consists of five stages, starting May 6, 2024. DTC tests are specifically noted as having never been subject to enforcement discretion, meaning compliance with FDA requirements has always been expected for consumer-facing diagnostic tests. However, on April 17, 2025, a federal court blocked FDA's final rule on LDTs, creating uncertainty about the regulatory future. == Degradation == Once secreted, GLP-1 is extremely susceptible to the catalytic activity of the proteolytic enzyme dipeptidyl peptidase-4 (DPP-4). Specifically, DPP-4 cleaves the peptide bond between Ala8-Glu9 resulting in the abundant GLP-1 (9–36) amide constituting 60–80% of total GLP-1 in circulation. DPP-4 is widely expressed in multiple tissues and cell types and exists in both a membrane-anchored and soluble circulating form. Notably, DPP-4 is expressed on the surface of endothelial cells, including those located directly adjacent to GLP-1 secretion sites. Consequently, less than 25% of secreted GLP-1 is estimated to leave the gut intact. Additionally, presumably due to the high concentration of DPP-4 found on hepatocytes, 40–50% of the remaining active GLP-1 is degraded across the liver. Thus, due to the activity of DPP-4 only 10–15% of secreted GLP-1 reaches circulation intact. Neutral endopeptidase 24.11 (NEP 24.11) is a membrane-bound zinc metallopeptidase widely expressed in several tissues, but found in particularly high concentrations in the kidneys, which is also identified accountable for the rapid degradation of GLP-1. It primarily cleaves peptides at the N-terminal side of aromatic or hydrophobic amino acids and is estimated to contribute by up to 50% to GLP-1 degradation.
