A documented effect of tesamorelin is a reduction in visceral adipose tissue in some study populations. Researchers have reported decreases in trunk fat measured by computed tomography alongside changes in lipid markers. The mechanism is thought to involve growth hormone-mediated lipolysis, though the precise contribution of direct versus indirect pathways is not fully resolved. Studies have generally examined defined groups over finite periods, so long-term outcomes are less well characterized. Findings have not been uniform across all trials.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone (GHRH). Its sequence corresponds to the 44-amino-acid form of human GHRH with a trans-3-hexenoyl group attached to the N-terminal tyrosine. This modification slows enzymatic cleavage and extends the peptide's activity relative to the native hormone. The compound is produced by solid-phase peptide synthesis and supplied as a lyophilized powder. Researchers classify it as a GHRH receptor agonist. Its structure places it in the same family as other growth hormone secretagogues that act on the pituitary.
Measured responses usually involve growth hormone and insulin-like growth factor 1, known as IGF-1. Growth hormone rises in bursts and is difficult to sample reliably, while IGF-1 shifts more slowly and can be assessed from a single blood draw. Studies therefore treat IGF-1 as the more practical pharmacodynamic marker. Both are indirect, showing that the receptor was engaged rather than that the peptide reached a particular concentration. Direct exposure measurement requires an assay aimed at the molecule itself.
Published work tends to frame tesamorelin as a tool for studying the GHRH axis and as a compound with measurable effects on body composition. Reports often describe visceral adipose tissue as an endpoint, assessed by imaging rather than by inference. Analytical sections commonly describe liquid chromatography with tandem mass spectrometry to confirm identity and purity, because immunoassays may cross-react with related fragments. Where results diverge between studies, differences in assay choice, sampling timing, and population are frequent explanations offered. Whether effects persist after treatment stops remains an open question.
Tesamorelin binds the growth hormone–releasing hormone receptor on pituitary somatotroph cells. The receptor signals through the Gs protein, raising intracellular cAMP and activating protein kinase A. That cascade triggers release of stored growth hormone in pulses rather than a steady stream. Because the drug acts at the receptor that normally controls this process, its effect depends on the body's own signaling architecture rather than on a synthetic pathway. The resulting hormone profile reflects the timing of each pulse, not only its size.

Clinical investigation has focused on HIV-associated lipodystrophy, a condition in which antiretroviral therapy contributes to abnormal fat distribution. Excess visceral adipose tissue accumulates in the abdomen while peripheral fat may be lost. Tesamorelin was evaluated for reducing this visceral fat depot, with trials measuring changes in abdominal fat by imaging rather than by body weight alone. The rationale rests on the known lipolytic effects of growth hormone. Effects on visceral fat are documented, while long-term outcomes regarding cardiovascular risk remain less clearly established.
Tesamorelin is a synthetic peptide analog of growth hormone-releasing hormone, composed of 44 amino acids. It was designed to retain the biological activity of the native hormone while resisting rapid enzymatic degradation. The compound is classified as a growth hormone secretagogue and belongs to the broader family of hypothalamic releasing factors. In research and clinical settings, it is studied for its ability to stimulate pituitary growth hormone release. Its structure includes a modification at the N-terminus that contributes to an extended half-life relative to native growth hormone-releasing hormone.
Pharmacodynamic studies show that tesamorelin reduces visceral adipose tissue more than subcutaneous adipose tissue in the studied population. This selectivity may relate to differences in blood flow and hormone sensitivity between fat depots. Effects on glucose metabolism and insulin sensitivity have been investigated, with some trials reporting modest changes and others showing stability. The precise relationship between growth hormone exposure, IGF-1 levels, and visceral fat loss remains an active area of analysis.
Tesamorelin binds to growth hormone-releasing hormone receptors on somatotroph cells in the anterior pituitary. Receptor activation increases intracellular cyclic AMP and promotes synthesis and secretion of growth hormone. Because the peptide mimics endogenous GHRH, it amplifies the normal pulsatile release of growth hormone rather than providing exogenous growth hormone directly. This upstream action distinguishes tesamorelin from recombinant growth hormone preparations and from growth hormone secretagogues that act at different receptors.

In North America, the 323 saloon became the Protegé, while the 323 hatchback kept its name. There, and in the Philippines, base (SE/DX), 4WD and top-line LX models were available. The 1990 base model has the SE name and uses the B8 1.8 L SOHC engine that has 16 valves and hydraulic lifters. In 1991, the base model name was changed to "DX". 4WD models existed for the 1990 and 1991 model years, with the SOHC engine and rear disc brakes. The LX version of the Protegé included a 1.8 L DOHC 16-valve BP engine producing 125 hp (93 kW; 127 PS). LX models also have power windows and door locks and 14-inch wheels. Vented front and solid rear disc brakes, larger front brakes, larger clutch, equal-length driveshafts, dual outlet muffler, body-color door handles and mirrors, fold-down rear center armrest, driver's vanity mirror, and larger stabilizer bars. A sunroof and 14-inch aluminum alloy wheels were options on LX models. The Protegé nameplate was also applied for 323 saloon in Brazil, which was really similar to the Canadian market specification. However, only a fuel injected 1.8 L SOHC engine was offered there. BG models that were assembled in New Zealand came with glass headlights instead of plastic, and a 240 km/h speedometer. In Thailand, the 323 saloon was initially offered in a single unnamed trim with a carbureted 1.6 L SOHC engine as the sole option. The facelifted model was divided into two levels: the 1.3i and 1.6i LX, and the better-equipped 1.6i GLX. All engines featured fuel injection, and the latter came with a 4-speed automatic transmission as an option.

Coordinates were measured in grades on official French terrestrial ordnance charts from the French revolution well into the 20th century. 1 grade (or in modern symbology, 1 gon) = 0.9° or 0.01 right angle. One advantage of this measure is that the distance between latitude lines 0.01 gon apart at the equator is almost exactly 1 kilometre. It would be exactly 1 km if the original definition of 1 metre = 1⁄10,000 quarter-meridian had been adhered to. One disadvantage is that common angles like 30° and 60° are expressed by fractional values (33+1⁄3 and 66+2⁄3 respectively) so this "decimal" unit failed to displace the "sexagesimal" units equilateral-vertex – degree – minute – second invented by Babylonian astronomers.
=== Industry and legal actions === DuPont has used PFOA for over 50 years at its Washington Works plant. Area residents sued DuPont in August 2001 and claimed DuPont released PFOA in excess of their community guideline of 1 part per billion resulting in lower property values and increased risk of illness. The class was certified by Wood Circuit Court Judge George W. Hill. As part of the settlement, DuPont has paid for blood tests and health surveys of residents believed to be affected. Participants numbered 69,030 in the study, which was reviewed by three epidemiologists—the C8 Science Panel—to determine if any health effects are the likely result of exposure. On December 13, 2005, DuPont announced a settlement with the EPA in which DuPont would pay US$10.25 million in fines and an additional US$6.25 million for two supplemental environmental projects without any admission of liability. On September 30, 2008, Chief Judge Joseph R. Goodwin of the United States District Court for the Southern District of West Virginia denied the certification of a class of Parkersburg residents exposed to PFOA from DuPont's facility because they did not "show the common individual injuries needed to certify a class action". On September 28, 2009, Judge Goodwin dismissed the claims of those residents except for medical monitoring. By 2015, more than three thousand plaintiffs have filed personal-injury lawsuits against DuPont. In 2017, DuPont reached a $670.7 million cash settlement related to 3,550 personal injury lawsuits tied to PFOA contamination of drinking water in the Parkersburg area.

==== Defence procurement ==== The Defence budget was protected by a NATO commitment to increase defence spending by 3% per annum until 1986, but was still subjected to cuts in the proposed budget during Heseltine's tenure. Some senior military figures felt that Heseltine was obsessed with the minutiae of running the department rather than thinking strategically about defence priorities and procurement. Dwin Bramall recalled that Heseltine never showed an interest in the strategy papers he sent him. Thatcher was highly critical of him for failing to take a decision on the development of the Nimrod early-warning plane, on which £660 million was spent over a ten-year period, only for the project to be cancelled by his successor. Some accusations were raised (the Commons Select Committee on Defence thought him "vague and evasive" on the issue in 1985) that the accounts were being massaged to push costs into the period after 1986, when cuts would become inevitable. The journalist Hugo Young later recalled Heseltine briefing journalists confidentially that spending and funding could be reconciled until 1986, by which time he expected "to be gone". In Cabinet, Heseltine resented being kept out of economic debates and suspected he might be reshuffled to the job of Secretary of State for Northern Ireland as Jim Prior had been. He had tried to pursue a one-man industrial policy, as defence spent £17 billion per annum, 5% of UK GDP, half of it on procurement, and 90% of that in the UK, with 700,000 British jobs dependent on it.
== Properties == DTT is oxidized by air. So it is normally stored and handled under inert atmosphere to minimize oxidation. The rate of air-oxidation is slower at low temperatures. Oxidized DTT exhibits a strong absorbance peak at 280 nm. Since thiols are less nucleophilic than their conjugate bases, thiolates, DTT becomes a less potent nucleophile as the pH falls. (2S)-2-Amino-1,4-dimercaptobutane (dithiobutylamine or DTBA), a related dithiol reducing agent, somewhat overcomes this limitation of DTT. Tris(2-carboxyethyl)phosphine (TCEP) is an alternative reducing agent that is more stable and effective at low pH, but it is bulky and reduces cystines in folded proteins only slowly. DTT's half-life is 40 hours at pH 6.5 and 1.4 hours at pH 8.5 and 20 °C; its half-life decreases further as temperature increases. The presence of EDTA (ethylenediaminetetraacetic acid) to chelate divalent metal ions considerably extends the half-life of DTT in solution.
== Food additive == Diglycerides, generally in a mix with monoglycerides (E471), are common food additives largely used as emulsifiers. The values given in the nutritional labels for total fat, saturated fat, and trans fat do not include those present in mono- and diglycerides. They often are included in bakery products, beverages, ice cream, peanut butter, chewing gum, shortening, whipped toppings, margarine, confections, and some snack products, such as Pringles.
=== Hylomorphism in contemporary anthropology === Anthropologist Tim Ingold has critiqued the classical hylomorphic model in his article The Textility of Making (2010), arguing that form is not imposed on passive matter by an external agent but instead "emerges within the process of making". He describes this alternative as a morphogenetic approach, influenced by process philosophy and drawing on ideas from Gilles Deleuze and Félix Guattari. Ingold emphasizes the active participation of materials, proposing that making is a relational and temporal unfolding rather than the execution of a predefined design.
=== Deficiency === Low plasma magnesium (hypomagnesemia) is common: it is found in 2.5–15% of the general population. From 2005 to 2006, 48 percent of the United States population consumed less magnesium than recommended in the Dietary Reference Intake. Other causes are increased renal or gastrointestinal loss, an increased intracellular shift, and proton-pump inhibitor antacid therapy. Most are asymptomatic, but symptoms referable to neuromuscular, cardiovascular, and metabolic dysfunction may occur. Alcoholism is often associated with magnesium deficiency. Chronically low serum magnesium levels are associated with metabolic syndrome, diabetes mellitus type 2, fasciculation, and hypertension.

=== Reactivity === Computational efforts have been vital in explaining the thermodynamics and kinetics of these cycloaddition reactions which has played a vital role in continuing to improve the reaction. There are two methods for activating alkynes without sacrificing stability: decrease transition state energy or decrease reactant stability.
The RICE method is an frequently used mnemonic acronym used in the initial treatment of a soft tissue injury. It has been widely challenged in recent years in favor of other treatment recommendations.

Whisky from the northern area is milder because it is made using spring water for a "lighter flavoured, mossy (rather than peaty), with some seaweed, some nuts" characteristic. The national tourist board website says that the single malts from Islay vary by distillery, from "robust and smoky" to "lighter and sweeter". The Lowlands: According to Visit Scotland, the website of the national tourist board, this district covers "much of the Central Belt and the South of Scotland including Edinburgh & The Lothians, Glasgow & The Clyde Valley, the Kingdom of Fife, Ayrshire, Dumfries & Galloway and the Scottish Borders". There were 18 Lowlands distilleries in the region as of 2019, according to the website of the national tourist board, including some that opened quite recently. These include well-known companies such as Annandale, Auchentoshan, Bladnoch, Glenkinchie, and Ailsa Bay in the site of the Girvan distillery as well as Daftmill, Eden Mill, Kingsbarns and Rosebank. Region characteristics: soft and smooth, consisting of a floral nose with a sweet finish. Single malts from this area tend to be "lighter, sweet and [with] floral tones". Speyside: Speyside gets its name from the River Spey, which cuts through this region and provides water to many of the distilleries. Speyside has approximately 50 distilleries within its geographic boundaries and has officially been recognised as a region, distinct from the Highlands, since 2014.

It mirrors the 44-residue form of human growth hormone-releasing hormone. A hexenoyl group on the N-terminal tyrosine distinguishes it from the unmodified hormone. The change is intended to improve resistance to enzymatic breakdown.
The N-terminal modification reduces cleavage by circulating peptidases, so the peptide persists longer than native GHRH. That persistence is the main rationale for the synthetic design. Comparative half-life values in humans are reported in regulatory review documents rather than in general reference literature.
Reductions in visceral adipose tissue have been measured in controlled studies of defined populations. Whether the effect generalizes to other groups and persists after treatment stops is less clear. Longer-term outcome data remain limited.
It acts on the growth hormone–releasing hormone receptor, a Gs-coupled receptor found on pituitary somatotroph cells. Activation raises cAMP and prompts pulsatile hormone release.
